DEVELOPMENT OF A METHOD FOR DETERMINING THE AMOUNT OF WATER-SOLUBLE VITAMINS USING THE HPLC METHOD (USING THE EXAMPLE OF JUJUBE)

Abstract
In the article, the results of the research on the development of the methodology for determining the amount of water-soluble B1, B2, B3, B6, B9, B12, C and PP vitamins in the plant using the HPLC method are described in the article. Optimum parameters of conditions for drying, crushing, extraction of plant sample, preparing it for chromatographic analysis and performing analysis have been determined.
When the extraction process of hydrophilic vitamins from the plant sample lasted 20 minutes with an increase in temperature from 20 °C to 80 °C, a tendency for the amount of vitamins in the extract to increase to 60 °C was observed. Therefore, 60 °C was chosen as the optimal extraction temperature. At the same constant temperature, an increase in time from 10 to 40 minutes showed that the optimal extraction time was 20 minutes.
Vitamins in standard solutions and analyzed samples were determined using a high-performance liquid chromatograph LC-40 Nexera Lite equipped with a GIST C18 reversed-phase column (150 × 4.6 mm; 5 μm, Shimadzu) using a gradient of acetonitrile and 0.25% aqueous acetic acid as the mobile phase. Chromatographic detection was carried out at wavelengths of 265, 291 and 550 nm.
It was confirmed that the accuracy of the analysis methodology developed on the basis of mathematical processing of the obtained results is sufficiently high. It was shown that jujube fruit contains the most vitamins B6, B3, B2 and C, and vitamin B12 is not found.
